What is CJC-1295 DAC?

CJC-1295 DAC (molecular mass 3775.4, same CAS family 863288-34-0) carries a maleimidopropionic acid group at the C-terminus that forms a covalent bond with cysteine-34 of serum albumin. The non-DAC form, catalogued separately as CJC-1295 (mass 3367.9), lacks that group. How the reported mechanism works

GHRH receptor agonism

Once bound to albumin, the conjugate still activates the GRF receptor on the anterior pituitary, which the 2005 Endocrinology study demonstrated in rats using hGRF(1-29)-albumin bioconjugates.

What the drug affinity complex changes

The DAC does not change which receptor is engaged; it changes exposure over time. That is why the two forms answer different experimental questions: pulsatile stimulation versus sustained elevation, a distinction that matters in any model where the pattern of receptor activation is part of the biology.

The 2006 clinical pharmacology study in healthy adults, which reported prolonged elevation of growth hormone and IGF-I, used this DAC form. Development stopped short of an approved product. For laboratory work the useful pairing is DAC with non-DAC in the same design, since the peptide sequence is constant and only the exposure profile differs.
