FOXO4-DRI (Proxofim) is a synthetic retro-inverso analogue of FOXO4, incorporating D-amino acids for enhanced proteolytic resistance. It competitively disrupts the FOXO4-p53 interaction that maintains senescent cell viability. Research Highlights 1. Senescent Cell Elimination — Restores p53-mediated apoptosis in senescent cells while sparing healthy tissue.
2. Tissue Rejuvenation — Aged mice showed increased activity, improved cognition, and enhanced fur density. Restored functional integrity in kidneys, liver, and skin.
3. Pathological Scarring — Promoted apoptosis in keloid fibroblasts and reduced G0/G1 phase proliferation.
4. Insulin Signaling — FOXO proteins mediate insulin/IGF effects on metabolism and aging.
