Melanotan 2 (MT-2) is a cyclic heptapeptide analogue of human α-MSH, originally synthesized at the University of Arizona during the 1980s. It functions as a non-selective melanocortin receptor agonist, binding primarily to MC-1R and MC-4R receptors, with implications for pigmentation, sexual function, appetite control, and metabolic regulation.

Research Highlights

1. Melanocortin Receptor Activation

MT-2 engages multiple melanocortin receptor subtypes. Through MC-1R binding, it stimulates melanin production and skin pigmentation. MC-4R activation influences glucose and cholesterol metabolism, thermogenic responses, and behavioral patterns.

2. Autism Spectrum Research

In a maternal immune activation mouse model, seven-day MT-2 administration improved social behavioral metrics, suggesting potential modulation of oxytocin-related pathways.

3. Appetite and Weight Regulation

MC-4R receptor engagement appears to influence dietary preferences, with studies showing significantly higher fat consumption among MC-4R-deficient individuals.

4. Glycemic Modulation

MT-2 injection into the ventromedial hypothalamus increased glucose uptake in skeletal muscle, brown adipose tissue, and cardiac tissue.

5. Alcohol Intake Reduction

Preclinical studies demonstrated MT-2's capacity to reduce alcohol consumption. When combined with naltrexone, efficacy was enhanced more than sevenfold.

6. Erectile Function Research

In controlled studies, MT-2 induced clinically apparent erections in 8 of 10 participants with psychogenic erectile dysfunction.
